WR33 belongs to a large protein domain family with homologues in several eukaryotic genomes, Homo sapiens, Mus musculis and Drosophila melanogaster
None of the members of this strongly conserved protein domain family has a characterized biological function
The Bos Taurus homologue (38% identity) is characterized as “brain specific protein P25”
WR33-180 (190 residues long) was studied previously
- Residues 20-120 have largely alpha-helical structure (5 helices)
- Residues 125-180 adopt largely disordered structure
- Assignment process very difficult due to severe overlap
Several constructs were produced to get rid of the tail and make amenable working with it while keeping 3D structure.
- WR33-109, WR33-113, WR33-115, WR33-117
- Based on HSQC screening, WR33-115 was selected
- Fast and reliable way of mapping domains
